Titre The effects of acute transfer to freshwater on ion transporters of the pharyngeal cavity in European seabass (Dicentrarchus labrax) Type Article scientifique

Résumé Gene expression of key ion transporters (the Na+/K+-ATPase NKA, the Na+, K+-2Cl(-) cotransporter NKCC1, and CFTR) in the gills, opercular inner epithelium, and pseudobranch of European seabass juveniles (Dicentrarchus labrax) were studied after acute transfer up to 4days from seawater (SW) to freshwater (FW). The functional remodeling of these organs was also studied. Handling stress (SW to SW transfer) rapidly induced a transcript level decrease for the three ion transporters in the gills and operculum. NKA and CFTR relative expression level were stable, but in the pseudobranch, NKCC1 transcript levels increased (up to 2.4-fold). Transfer to FW induced even more organ-specific responses. In the gills, a 1.8-fold increase for NKA transcript levels occurs within 4days post transfer with also a general decrease for CFTR and NKCC1. In the operculum, transcript levels are only slightly modified. In the pseudobranch, there is a transient NKCC1 increase followed by 0.6-fold decrease and 0.8-fold CFTR decrease. FW transfer also induced a density decrease for the opercular ionocytes and goblet cells. Therefore, gills and operculum display similar trends in SW-fish but have different responses in FW-transferred fish. Also, the pseudobranch presents contrasting response both in SW and in FW, most probably due to the high density of a cell type that is morphologically and functionally different compared to the typical gill-type ionocyte. This pseudobranch-type ionocyte could be involved in blood acid-base regulation masking a minor osmotic regulatory capacity of this organ compared to the gills.

Résumé Biological variability is no longer considered as statistical noise, but rather as an adaptive benefit. This variability comes from consistent differences in behavioral and physiological responses among individuals to a changing/challenging environment, named “coping style”, “temperament” or “personality”. Many studies have described how to characterize personality traits and how to assess their consistency over time and between different contexts; however, little is known about the environmental factors shaping personality development. Because contrasting personalities are maintained with evolution, this lead to the widespread assumption that genes play a predominant role in personality. In many cases, personality traits are however also likely to be determined by individual experience, which is probably at least as important as genetics in shaping personality. The aim of this study was to assess how environmental variability (herein food predictability) impacts behavioral responses, particularly the shyness-boldness axis, one of the most widely shared animal personality trait. Here, we reared juvenile seabass (95 days old) from two divergent strains selected for feed deprivation tolerance under standard conditions for 40 days. Thereafter, we submitted them to two feeding treatments (Predictable versus Unpredictable) starting at 135 dph and lasting 60 days. Seabass reared under a predictable food supply (PFS) grew faster and were shyer than fish reared under an unpredictable food supply (UFS) (i.e. they took more time to exit the refuge zone of a Z-maze; UFS: 132.47 ± 34.63 s; PFS: 336.79 ± 56.97 s) but their exploration tendency was similar. We also examined the behavioral responses of these fish facing a hypoxic challenge. Hypoxia tolerance results were consistent before and after the two feeding treatments. Our findings show the importance of early environmental experience as a driving force shaping boldness. In addition, we provide further evidence that predictable feeding time should be respected in studies assessing essential functions such as growth and behavior. Although personality traits are partially heritable, this study demonstrates the important influence of environmental conditions and life history on behavior.

Titre Consistency in European seabass coping styles: A life history approach Type Article scientifique

Résumé Recent years have seen a growth of interest in the consistent differences in individual behaviour over time and contexts constituting so-called “individual coping styles”. An understanding of this inter-individual variation is essential to improve our knowledge of the adaptive value of behaviour. Coping styles may have implications in diverse fields, so the development of appropriate screening methods for each species appears to be the most effective way to extend our knowledge and to incorporate behavioural responses into selection-based breeding programmes, to improve the domestication and welfare of farmed fish. We tested 30 juvenile seabass (Dicentrarchus labrax) at least twice in individual-based tests (feeding recovery in isolation, aggressiveness, exploration in a T-maze and net restraint) and group-based tests (risk-taking and hypoxia sorting), to assess coping style consistency in the short and long term and between tests. The results of individual-based tests were inconsistent over time and between tests in our set-up: the time between repeat tests, learning and species-specific behavioural responses appeared to have a major impact. By contrast, the results of group-based tests, such as risk-taking and hypoxia sorting, appeared to be consistent (both in the short and long term). These tests therefore appeared to be the most relevant for the characterisation of coping style in European seabass. Furthermore, the results of these tests were also predictive of cortisol stress response. These tests are simple to perform and can be used to screen large numbers of fish, the first step in selection programmes including behavioural profiles

Résumé Supplies of marine fish oils are limited, and continued growth in aquaculture production dictates that lipid substitutes in fish diets must be used without compromising fish health and product quality. In this study, the total substitution of a fish meal and fish oil by a blend of vegetable meals (corn, soybean, wheat and lupin) and linseed oil in the diet of European sea bass (Dicentrachus labrax) was investigated. Two groups of European sea bass were fed with fish diet (FD) or vegetable diet (VD) for 9 months. VD, totally deprived of eicosapentaenoate (EPA; 20:5n-3) and docosahexaenoate (DHA; 22:6n-3), revealed a nutritional deficiency and affected growth performance. Whilst VD induced a significant increase in fatty acid desaturase 2 (FADS2) and sterol binding regulatory element-binding protein 1 (SREBP-1) mRNA levels, the desaturation rate of [1-C-14]18:3n-3 into [1-C-14]18:4n-3, analysed in microsomal preparations using HPLC method, did not show an upregulation of FADS2 activities in liver and intestine of fish fed VD. Moreover Western-blot analysis did not revealed any significant difference of FADS2 protein amount between the two dietary groups. These data demonstrate that sea bass exhibits a desaturase (FADS2) activity whatever their diet, but a post-transcriptional regulation of fads2 RNA prevents an increase of enzyme in fish fed a HUFA-free diet. This led to a lower fish growth and poor muscle HUFA content.
